This chapter will provide an up-to-date depiction of the molecular networks involved in the osteogenic process, focusing on main genes and signaling pathways (schematically represented in Figure 1), whose integrity is required for the correct skeletal morphogenesis and patterning and for maintaining bone homeostasis. Particular attention will be devoted to list and dissect the human syndromes and disorders associated to genes belonging to the main osteogenic pathways, with regard to the skeletal phenotype
